Specificity and Use

CENPU / MLF1IP antibody was raised against synthetic peptide from human MLF1IP / PBIP1.
pThr78
Amino acids surrounding Thr78 of human MLF1IP protein. The immunogen peptide is phosphorylated at Thr78.
Immunohistochemistry: LS-B71 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B71 was determined to be 20 ug/ml.

Packaging

0.02 M potassium phosphate, 0.15 M sodium chloride, pH 7.2, 0.01% sodium azide.
Store vial at -20 C prior to opening. Dilute only prior to immediate use. For extended storage, aliquot contents and freeze at -20 C or below. Avoid cycles of freezing and thawing.

Myeloid leukemia factor-1 (MLF1) Interacting protein, also known as PBIP1, is a novel gene that encodes a putative transcriptional repressor. It is localized to human chromosome 4q35.1, is expressed in both the nuclei and cytoplasm of cells, and is a novel substrate for polo-like kinase (PLK1). Phosphorylation by PLK1 regulates the PLK1-MLF1 interaction and localization to the kinetochores promoting proper chromosome segregation.

Anti-MLF1IP pT78 Antibody - Western Blot. Western blot of affinity purified anti-MLF1IP pT78 antibody shows detection of MLF1IP phosphorylated at Thr78. HeLa cells were co-infected with the indicated adenoviruses expressing GFP-tagged Plk1 or PBIP1. Blots were probed with the anti-MLF1IP pT78 antibody, stripped, and then reprobed with anti-GFP antibody (Kang & Park, et al., 2006).
