Western blot using the affinity purified anti-PAI-2 antibody shows detection of endogenous PAI-2 in 50 µg of HeLa whole cell lysates. The band at ~42 kDa (arrowhead) corresponds to PAI-2. Faint non-specific bands are also noted at lower and higher molecular weights positions. Primary antibody was used at a 1:500 dilution in 5% BLOTTO in PBS reacted overnight at 4°C. The membrane was washed and reacted with a 1:5,000 dilution of HRP- conjugated Gt-a-Rabbit IgG for 45 min at room temperature with ECL used for detection. Molecular weight estimation was made by comparison to prestained MW markers.
