Specificity and Use

BAP / SIL1 antibody was raised against synthetic peptide C-ELLGSVNSLLKELR from the C-terminus of human SIL1 (NP_071909.1; NP_001032722.1). Percent identity by BLAST analysis: Human, Gorilla, Monkey (100%); Marmoset, Dog, Bat, Horse (93%); Hamster, Panda, Rabbit (86%).

Human SIL1. Variants (NP_071909.1; NP_001032722.1) encode the same isoform.
Immunohistochemistry: LS-B3286 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B3286 was determined to be 5 ug/ml.

BAP / SIL1 is a resident endoplasmic reticulum (ER), N-linked glycoprotein with an N-terminal ER targeting sequence, 2 putative N-glycosylation sites, and a C-terminal ER retention signal. This protein functions as a nucleotide exchange factor for another unfolded protein response protein. Mutations in this gene have been associated with Marinesco-Sjogren syndrome. Alternate transcriptional splice variants have been characterized.
