Specificity and Use

NS0-derived rhCathepsin S (aa 17 - 331).
Human Cathepsin S.
Western blot-This antibody can be used at 0.1-0.2 µg/ml with the appropriate secondary reagents to detect human pro and active Cathepsin S. The detection limit for rhCathepsin S is approximately 25 ng/lane under non-reducing and reducing conditions. Immunoprecipitation-This antibody has been used at 25 µg/ml to immunoprecipitate rhCathepsin S from conditioned media of cell culture. Immunohistochemistry-This antibody can be used to detect Cathepsin S in cells and tissues. The recommended working concentration is 5-15 pg/ml. ELISA capture-This product can be used as a capture reagent in a human Cathepsin S sandwich immunoassay in combination with biotinylated human. Cathepsin S detection antibody and recombinant human Cathepsin S as the standard. The suggested coating concentration range is. 0.2-0.8 µg/ml and should be titrated to determine the optimal concentration. In this format, less than 0.05% cross-reactivity with rhCathepsin A, -B, -C, -D, E, L, V, and -X/Z/P was observed. Neutralization-This antibody has been used to neutralize the enzymatic activity of rhCathepsin S. The antibody was preincubated with the activated enzyme at different molar ratios at room temperature for 30 minutes. The enzyme was then assayed with a peptide substrate (7-methoxycoumarin4-yl)acetyl-Arg-Pro-Lys-Pro-Val-Glu-Nva-Trp-Arg-Lys (2, 4-dinitrophenyl)-NH2. 50% of the proteolytic activity was inhibited by the antibody at approximately 1.23 µg/ml (IC50) under conditions in which the enzyme was present at 0.25 µg/ml and the substrate concentration was 10 pM. Considering the molecular masses of the enzyme (30 kD) and the antibody (150 kD), IC50 was achieved at approximately 1:1 molar ratio of the antibody to the enzyme.
