Specificity and Use

CTSD / Cathepsin D antibody was raised against cathepsin D purified from human spleen.
Detects cathepsin B, a lysosomal cysteine protease which functions as both a endopeptidase and a carboxypeptidase. Together with cathepsins D and L, cathepsin B is one of the most abundant proteases in the lysosome, and participates in intracellular protein degradation. Cathepsin B is thought to play an increasingly important role in apoptosis and its release from the lysosome under cell stress leads to Bid cleavage, cytochrome C release and nuclear fragmentation. Cathepsin B is also known as amyloid precursor protein secretase and is involved in the proteolytic processing of amyloid precursor protein (APP). Incomplete processing of APP is thought to be a causative factor in Alzheimer disease. Over-expression of cathepsin B has been associated with esophageal adenocarcinoma and other tumors, and has in particular been implicated in tumor invasion and metastasis.
Immunohistology: Trypsinization may be required.

CTSD / Cathepsin D is a lysosomal aspartyl protease composed of a dimer of disulfide-linked heavy and light chains, both produced from a single protein precursor. This proteinase, which is a member of the peptidase C1 family, has a specificity similar to but narrower than that of pepsin A. Transcription of this gene is initiated from several sites, including one which is a start site for an estrogen-regulated transcript.
