Specificity and Use

Collagen VII antibody was raised against insoluble fractions of human neonatal foreskin epidermal cells. MW of Antigen: 250kD (Intact); 150kD (Digested).
Recognizes intact (250kD) and collagenase digested (150kD) tissue Type VII collagen. Does not cross-react with types I, II, III, IV, V collagens, lamins and fibronectin. Epitope: Located on non-helical (non-collagenous) C-terminal region of collagen VII and is resistant to collagenase, trypsin, pronase, neuraminidase, chondroitinase, and hyaluronidase However, it is lost by digestion of collagen VII with pepsin. Species cross-reactivity: human, marmoset, monkey, bovine, sheep, goat, porcine and guinea pig.
Suitable for use in Electron Microscopy, Immunofluorescence and Immunohistochemistry. Immunohistochemistry: Acetone-fixed frozen only. Fixation in methanol is not suitable. Positive control: Normal skin.

About Collagen VII

The type VII collagen fibril, composed of three identical alpha collagen chains, is restricted to the basement zone beneath stratified squamous epithelia. It functions as an anchoring fibril between the external epithelia and the underlying stroma. Mutations in this gene are associated with all forms of dystrophic epidermolysis bullosa.
