Specificity and Use
CASP8 / Caspase 8 antibody was raised against recombinant full-length human Caspase 8 protein.
Reacts with a 55 kD protein. Caspase 8 has an N-terminal domain with sequence homology to the death effector domain (DED) of FADD. Caspase 8 binds to DED of FADD. Caspase 8 is activated by association with Fas/FADD death-inducing signaling complex to release two active subunits, p18 and p10 into the cytosol, where they activate other Caspases amplifying the apoptotic signal. This antibody cross reacts with mouse, rat, monkey and cow.
Most upstream protease of the activation cascade of caspases responsible for the TNFRSF6/FAS mediated and TNFRSF1A induced cell death. Binding to the adapter molecule FADD recruits it to either receptor. The resulting aggregate called death-inducing signaling complex (DISC) performs CASP8 proteolytic activation. The active dimeric enzyme is then liberated from the DISC and free to activate downstream apoptotic proteases.
