Specificity and Use

AXIN1 / Axin-1 antibody was raised against kLH-conjugated, synthetic peptide corresponding to amino acids 898-912 (SQRKAGGGSAPPCDS) of mouse axin. The immunizing sequence is identical in rat and has 13/15 identical amino acids in human. Percent identity by BLAST analysis: Mouse, Rat, Hamster (100%); Gibbon, Baboon, Monkey (93%); Human, Gorilla, Elephant, Dog, Bovine (87%); Marmoset, Panda, Bat (80%).
Recognizes recombinant mouse Myc-tagged Axin expressed in transfected cell lysate, Mr 130kD. Predicted to cross-react with rat and human based on sequence homology.
Western Blot: 1-2 ug/ml detects Axin in RIPA lysates from Cos-1 cells transfected with full length, mouse axin. Lysates of Cos-1 cells untransfected (lane 1) and expressing transfected Axin (lane 2) were resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Axin (1 ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.

Component of the beta-catenin destruction complex required for regulating CTNNB1 levels through phosphorylation and ubiquitination, and modulating Wnt-signaling. Controls dorsoventral patterning via two opposing effects; down-regulates CTNNB1 to inhibit the Wnt signaling pathway and ventralize embryos, but also dorsalizes embryos by activating a Wnt-independent JNK signaling pathway. In Wnt signaling, probably facilitates the phosphorylation of CTNNB1 and APC by GSK3B.
