Specificity and Use

ALPI / Alkaline Phosphatase antibody was raised against purified alkaline phosphatase from human intestine
Alkaline Phosphatase [Human Intestine].
Immunohistochemistry: LS-B326 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B326 was determined to be 2.5 ug/ml.

There are at least four distinct but related alkaline phosphatases: intestinal, placental, placental-like, and liver/bone/kidney (tissue non-specific). The intestinal alkaline phosphatase gene encodes a digestive brush-border enzyme. This enzyme is upregulated during small intestinal epithelial cell differentiation.

Anti-Alkaline Phosphatase Antibody - Western Blot. Both the antiserum and IgG fractions of anti-Alkaline Phosphatase (Human Intestine) are shown to detect under reducing conditions of SDS-PAGE the 60000 dalton enzyme in cellular extracts. Approximately 10 ug of total protein is loaded per lane. A 1:5000 dilution of the primary antibody is used followed by detection using HRP Goat-a-Rabbit IgG [H&L] (LS-C60884) diluted 1:4000 and color development using 4-CN substrate until sufficient color develops. Other detection systems will yield similar results.
