Specificity and Use

AIFM1 / AIF / PDCD8 antibody was raised against kLH-conjugated, synthetic peptide corresponding to amino acids 593-606 of human Apoptosis Inducing Factor, AIF (C-KDGEQHEDLNEV-AK), containing a N-terminal cysteine. The immunizing sequence is identical in mouse and rat. Percent identity by BLAST analysis: Human, Gorilla, Gibbon, Monkey, Marmoset, Mouse, Rat, Hamster, Elephant, Panda, Bovine, Dog, Bat, Cat, Horse, Rabbit, Pig (100%); Opossum, Xenopus (92%); Turkey, Chicken, Stickleback, Pufferfish, Zebrafish (83%).

Recognizes AIF, Mr 57kD. Another non-specific protein was also detected at Mr 120kD. Species cross-reactivity: Mouse. Predicted for human and rat based on sequence homology.
Suitable for use in Western Blot. Western Blot Analysis: 1 ug/ml detects AIF from a mouse liver preparation. Mouse liver protein preparation was resolved by electrophoresis, transferred to nitrocellulose and probed with AIF (1 ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.

Apoptosis is characterized by several morphological nuclear changes including chromatin condensation and nuclear fragmentation. These changes are triggered by the activation of members of caspase family, caspase activated DNase, and several novel proteins. A novel gene, the product of which causes chromatin condensation and DNA fragmentation, was recently identified, cloned, and designated apoptosis inducing factor (AIF).
