Immunohistochemical analysis of AARS2 staining in human liver cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with hematoxylin and mounted with DPX.

AARS2 antibody was raised against synthetic peptide C-QHRARQAEPVQK from an internal region of human AARS2 / AlaRS (NP_065796.1). Percent identity by BLAST analysis: Human, Chimpanzee, Gorilla (100%); Orangutan, Monkey (92%); Bovine, Porcine (83%).

Human AARS2 / AlaRS.
Peptide ELISA: antibody detection limit dilution 1:32000. Western blot: In transfected 143B transiently expressing Human AARS2 a band of approx. 100kD is observed. This band is barely visible in the non-transfected 143B. The calculated molecular size is 107kD according to NP_065796.1. Recommended concentration, 1-3 ug/ml. An additional non-specific 26kD was observed. We call for caution when used for other assays than Western blot.

The protein encoded by this gene belongs to the class-II aminoacyl-tRNA synthetase family. Aminoacyl-tRNA synthetases play critical roles in mRNA translation by charging tRNAs with their cognate amino acids. The encoded protein is a mitochondrial enzyme that specifically aminoacylates alanyl-tRNA. Mutations in this gene are a cause of combined oxidative phosphorylation deficiency 8.
