V5 Tag

V5 Tag, GKPIPNPLLGLDST, is from the C-terminal sequence of the P and V proteins of Simian Virus 5. V5 tag antibodies are used in many applications such as western blot, immunoprecipitation and immunofluorescence, and are a reliable means of purifying target proteins in many species. The V5 tag is often used alongside His-tags in affinity purification (PMID 24510596).

V5 Tag Antibody - Anti-V5 Epitope Tag Polyclonal Antibody - Western Blot. Anti-V5 epitope tag polyclonal antibody detects V5-tagged recombinant protein by western blot. This antibody was used at 1.0 ug/ml to detect 0.05 ug (lane 2) of full-length recombinant mouse serum albumin containing the V5 epitope tag at the carboxy end. Comparison to MW markers (lane 1) indicates detection of monomeric V5 tagged albumin. A 4-20% gradient gel was used to separate the protein by SDS-PAGE under non-reducing conditions. The protein was transferred to nitrocellulose using standard methods. After blocking the membrane was probed with the primary antibody overnight at 4C followed by washes and reaction with a 1:10000 dilution of IRDye 800 conjugated Gt-a-Rabbit IgG [H&L] (code for 45 min at room temperature. LICORs Odyssey Infrared Imaging System was used to scan and process the image. Other detection systems will yield similar results.

V5 Tag Antibody - Immunofluorescent analysis of V5-tag staining in 293T cells transfected with a V5-tag protein. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 deg C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
