PTPD1, a NT6 non-receptor protein tyrosine phosphatase, contains a N-terminal domain that is similar to cytoskeletal-associated proteins of the band 4.1-ezrin-radixin-moesin family and a single PTP domain at the C terminus. In transient coexpression experiments PTPD1 has been shown to be efficiently phosphorylated by and associated with the src kinase pp60src. Additionally, it has been shown to interact with BMX/ETK, a member of Tec tyrosine kinase family, and this interaction has been found to modulate the activation of STAT3 kinase. PTPD1 has been shown to affect liver regeneration and spermatogenesis in mice. At least two splice variants have been reported. The 2.9kb transcript has been shown to possess the catalytic domain but not the domains that associate with Src kinase. PTPD1 has been shown to be overexpressed in microsatellite instability pathway (MIN)-type sporadic colorectal cancer.
