ESRP2 / RBM35B

epithelial splicing regulatory protein 2

mRNA splicing factor that regulates the formation of epithelial cell-specific isoforms. Specifically regulates the expression of FGFR2-IIIb, an epithelial cell-specific isoform of FGFR2. Also regulates the splicing of CD44, CTNND1, ENAH, 3 transcripts that undergo changes in splicing during the epithelial-to-mesenchymal transition (EMT). Acts by directly binding specific sequences in mRNAs. Binds the GU-rich sequence motifs in the ISE/ISS-3, a cis-element regulatory region present in the mRNA of FGFR2.

ESRP2 / RBM35B Antibody - Anti-ESRP2 by western blot shows detection of ESRP2 in transfected 293T cell extracts (lane 2). Lanes ug and 1 contain 5 ug GFP-transfected- and ESRP1-transfected 293T cell lysates, respectively. Briefly, each lane contains approximately 5 ug of lysate. Primary antibody was used at a 1:1000 dilution (PBS-T plus milk) and reacted for O/N at 4C. The membrane was washed and reacted with a 1:10000 dilution of an anti-mouse ECL antibody for 1hr at room temperature. The bands shown are full length FLAG-ESRP2 (~80kDa) and a slightly lower band that is specific to ESRP2. Molecular weight estimation was made by comparison to prestained MW markers.

ESRP2 / RBM35B Antibody - Immunohistochemistry image at a dilution of 1:300 and staining in paraffin-embedded human breast cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0) . Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4 °C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
