ATP6V1D / V-ATPase subunit D is a component of vacuolar ATPase (V-ATPase), a multisubunit enzyme that mediates acidification of eukaryotic intracellular organelles. V-ATPase dependent organelle acidification is necessary for such intracellular processes as protein sorting, zymogen activation, receptor-mediated endocytosis, and synaptic vesicle proton gradient generation. V-ATPase is composed of a cytosolic V1 domain and a transmembrane V0 domain. The V1 domain consists of three A and three B subunits, two G subunits plus the C, D, E, F, and H subunits. The V1 domain contains the ATP catalytic site. The V0 domain consists of five different subunits: a, c, c', c", and d. Additional isoforms of many of the V1 and V0 subunit proteins are encoded by multiple genes or alternatively spliced transcript variants. This gene encodes the V1 domain D subunit protein.

ATP6V1D / V-ATPase subunit D Antibody - Staining HepG2 cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 min at 25°C. The primary antibody was diluted at 1:200 and incubated with the sample for 1 hour at 37°C. An Alexa Fluor 594 conjugated goat anti-rabbit IgG (H+L) antibody, diluted at 1/600, was used as secondary antibody.

The secondary antibody used was an HRP Goat Anti-Rabbit IgG (H+L) at 1:10000 dilution. Lysates were loaded 25ug per lane and 3% nonfat dry milk in TBST was used for blocking. An ECL Kit was used for detection and the exposure time was 60s.
