Usage Summary
Biological activity test: 1. Prepare a 180ul reaction buffer into a suitable container: Test is 120ul of 200mM Tris-HCl, pH8.0, 50ul of 20nM chymotrypsin and 10ul of 50ug/ml FKBP4. Blank is 130ul of 200mM Tris-HCl, pH8.0, 50ul of 20nM chymotrypsin. 2. Load 20ul of 5mM suc-AAFP-pNA into plate. (Substrate was dissolved in TFE that cantained 460mM LiCl to a concentration of 3mM) 3. Add prepared reaction buffer 180ul. 4. Record the increase in 405nm for 40 minutes at 37?.
Bio-Activity
Specific activity is > 700 nmol/min/mg, and is defined as the amount of cleaves 1nmole of suc-AAFP-PNA per minute at 37C in Tris-HCl pH 8.0 using chymotrypsin. Binding to FK506 has not been tested.
