1. Measured by its binding ability in a functional ELISA. Immobilized human TNFRSF10D Fc Chimera at 10 µg/ml (100 µl/well) can bind biotinylated TNFSF10 with a linear range of 0.625-20 ng/ml. 2. Measured by its ability to inhibit TRAIL-mediated cytotoxicity using L-929 mouse fibroblast cells treated with TRAIL. The ED50 for this effect is typically 0.2-1 µg/mL in the presence of 20 ng/ml Recombinant Human TRAIL/TNFSF10.

Apoptosis is induced by certain cytokines including TNF and Fas ligand in the TNF family through their death domain containing receptors. TRAIL/Apo2L is a new member of the TNF family and induces apoptosis of a variety of tumor cell lines. DR4 and DR5 are the recently identified functional receptors for TRAIL, and DcR1/TRID is a decoy receptor. Another member of the TRAIL receptor family was more recently identified and designated DcR2, TRAIL-R4, or TRUNDD.
