NAD-dependent protein deacetylase, which deacetylates internal lysines on histone and alpha-tubulin as well as many other proteins such as key transcription factors. Participates in the modulation of multiple and diverse biological processes such as cell cycle control, genomic integrity, microtubule dynamics, cell differentiation, metabolic networks, and autophagy. Plays a major role in the control of cell cycle progression and genomic stability.
HDAC activity (HDAC Fluorimetric Assay -AK-500 from ELS). This assay is performed in two stages. In the first stage, the HDAC(s) acts on the Substrate (10 uM, 50 uM) (total volume of 50 ul). The second stage, is initiated by the addition of 50 ul of developer, stops HDAC activity and produces the fluorescent signal. The samples are read in a microplate reading fluorimeter, capable of excitation at a wavelength in the range 350~380nm and detection of emission light in the range 440~460nm.
