Biological activity test: 1. Prepare a 180ul reaction buffer into a suitable container: Test is 120ul of 200mM Tris-HCl, pH8.0, 50ul of 20nM chymotrypsin and 10ul of 50ug/ml FKBP4. Blank is 130ul of 200mM Tris-HCl, pH8.0, 50ul of 20nM chymotrypsin.
2. Load 20ul of 5mM suc-AAFP-pNA into plate. (Substrate was dissolved in TFE that cantained 460mM LiCl to a concentration of 3mM)
3. Add prepared reaction buffer 180ul.
4. Record the increase in 405nm for 40 minutes at 37?.
FKBP4 / FKBP52 is a member of the immunophilin protein family, which play a role in immunoregulation and basic cellular processes involving protein folding and trafficking. This encoded protein is a cis-trans prolyl isomerase that binds to the immunosuppressants FK506 and rapamycin. It has high structural and functional similarity to FK506-binding protein 1A (FKBP1A), but unlike FKBP1A, this protein does not have immunosuppressant activity when complexed with FK506.
