Description
Tyrosinase (TYR) is an enzyme that catalyzes steps in the conversion of tyrosine to melanin. Mutations in TYR results in oculocutaneous albinism, and polymorphisms results in variations in skin pigmentation. The protein is found in melanocytes in the skin and retina, and is useful to identify melanocytic neoplasms such as melanomas. It is also found in subsets of renal angiomyolipomas and PEComas.

Specificity
Recognizes a cluster of proteins between 70-80kD, identified as human tyrosinase. Occasionally a minor band at 55kD is also detected. Does not cross-react with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Species cross-reactivity: canine and feline. Weak cross-reactivity with mouse.

Usage
Immunohistochemistry: LS-B2473 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2473 was determined to be 10 ug/ml.
