Specifications

Description
PDPN (podoplanin) is mucin-type membrane glycoprotein with expression in a diverse range of tissues and cell types including lung alveolar cells, kidney podocytes, lymphatic endothelial cells, and neural tissues. PDPN is a required protein in the development of the lungs, lymphatic system and the heart, and it separately may play a role in immune response to fungal infections. In immunohistochemistry, PDPN has cytoplasmic and membranous staining in the lymphatic endothelium, breast myoepithelium, podocytes, dendritic cells and mesothelium. It is a sensitive and specific marker for the lymphatic endothelium, and it is used as an endothelial cell marker in determining lymphatic invasion in tumors. Usage
Dilute antibody to its recommended working dilution using the same buffer defined in the Presentation. Manual IHC Protocol: 1) Perform Heat-Induced Epitope Retrieval at pH 9 for 10 to 30 minutes. 2) Block in peroxidase blocking solution for 5 minutes at room temperature. (Not required if using Alkaline Phosphatase System) 3) Dilute antibody 1:100-1:200 before applying. Incubate antibody for 10 to 30 minutes at room temperature. 4) Add secondary antibody and incubate for 20 to 30 minutes at room temperature. 5) Add DAB or Fast Red and incubate for 5 to 10 minutes at room temperature. 6) Counterstain with hematoxylin for 0.5 to 5 minutes, depending on the hematoxylin used. Rinse with distilled water and blueing solution for 30 seconds. Dehydrate and apply coverslip.
