Description
TFE3 (Transcription factor binding to IGHM enhancer 3) is a nuclear protein that promotes the expression of genes downstream to TGF-beta signaling. Chromosomal translocations between this gene (located on Xp11.2) and other genes promote growth and proliferation, and oncogenic activation. Translocation or overexpression of this gene has been implicated in renal cell carcinoma, lung adenocarcinoma, alveolar soft-part sarcoma, papillary thyroid carcinoma, melanoma, and mesotheliomas. It is expressed in a wide variety of tissues, and is nuclear in localization.

Usage
Dilute antibody to its recommended working dilution using the same buffer defined in the Presentation. Manual IHC Protocol: 1) Perform Heat-Induced Epitope Retrieval at pH 9 for 10 to 30 minutes. 2) Block in peroxidase blocking solution for 5 minutes at room temperature. (Not required if using Alkaline Phosphatase System) 3) Dilute antibody 1:100-1:200 before applying. Incubate antibody for 10 to 30 minutes at room temperature. 4) Add secondary antibody and incubate for 20 to 30 minutes at room temperature. 5) Add DAB or Fast Red and incubate for 5 to 10 minutes at room temperature. 6) Counterstain with hematoxylin for 0.5 to 5 minutes, depending on the hematoxylin used. Rinse with distilled water and blueing solution for 30 seconds. Dehydrate and apply coverslip.
