MRE11A / MRE11 Antibody - Anti-Mre11 Antibody - Western Blot. Western blot of Affinity Purified anti-Mre11 antibody shows detection of a band ~80 kD corresponding to mouse Mre11 (arrowhead). Lanes 1-4 contain 0.5 ug, 0.3 ug, 0.1 ug and 0.05 ug of purified mouse Mre11 protein, respectively. After 4-20% SDS-PAGE and transfer onto nitrocellulose, the membrane was blocked and then probed with the primary antibody diluted to 1:1000 overnight at 4C. The membrane was then washed and reacted with a 1:10000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX ( for 45 min at room temperature. IRDye800 fluorescence image was captured using the Odyssey Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.

Description
MRE11A is a component of the MRN complex, which plays a central role in double-strand break (DSB) repair, DNA recombination, maintenance of telomere integrity and meiosis. MRE11A provides the single-strand endonuclease activity and double-strand-specific 3'-5' exonuclease activity for this complex. MRE11A functions in microhomology-mediated end joining repair (MMEJ) of double strand breaks, which is an error-prone process. MRE11A is overexpressed in breast cancers, where it is thought to promote proliferation via STAT3, invasion and migration, and where increased repair functions may contribute to tumor cell radioresistance (and where it may alternatively contribute to the mutation burden). Furthermore, inherited polymorphisms in MRE11A are associated with risk for breast and ovarian cancer. Additionally, a study by Sedghi demonstrated a connection between MRE11A and an ataxia-telengiectasia-like disorder; a family of individuals that lack MRE11A expression showed a predisposition for developmental delay, intellectual disability, limb ataxia and cerebellar atrophy. This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a recombinant protein corresponding to amino acids 68-706 of mouse Mre11 protein.
Specificity
A BLAST analysis was used to suggest cross reactivity with Mre11 protein from rat (94% homology). Based on protein sequence homology also expect partial reactivity against Mre11 homologues from macaque (88%), human (84%) and dog (82%). This sequence shows less than 75% homology to homologues from bovine, Xenopus, chicken, chimpanzee and zebrafish sources. Reactivity against homologues from other sources is not known.

Usage
Immunohistochemistry: LS-B1447 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B1447 was determined to be 5 ug/ml.
