HP / Haptoglobin Antibody - Anti-Haptoglobin antibody IHC of human liver. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody concentration 31 ug/ml.
HP / Haptoglobin Antibody - ELISA results of purified Rabbit anti-Human Haptoglobin Antibody tested against immunizing antigen. Each well was coated in duplicate with 1.0 µg of antigen. The starting dilution of antibody was 5µg/ml and the X-axis represents the Log10 of a 3-fold dilution. This titration is a 4-parameter curve fit where the IC50 is defined as the titer of the antibody. Assay performed using 3% fish gelatin as blocking buffer, Goat anti-Rabbit IgG Antibody Peroxidase Conjugated (Min X Bv Ch Gt GP Ham Hs Hu Ms Rt & Sh Serum Proteins) and TMB substrate

Specifications

Description
HP (Haptoglobin) is a protein produced by hepatic cells that binds free plasma hemoglobin to make it accessible to degradative enzymes. It serves a protective function in the kidneys by preventing the loss of iron and protecting them from hemoglobin-induced damage. It has also been shown to have antimicrobial activity against bacteria. HP mutations and regulatory defects cause ahaptoglobinemia and hypohaptoglobinemia. Inherited HP polymorphisms are also associated with diabetic nephropathy, the incidence of coronary artery disease in type 1 diabetes, inflammatory disease behavior, Crohn's disease, primary sclerosing cholangitis, and susceptibility to idiopathic Parkinson's disease. In immunohistochemistry, HP has cytoplasmic and secretory positivity in the liver, and is also found in the kidneys, skin, lung and adipose tissue. References: The UniProt Consortium. Immunogen
Human Haptoglobin Antibody was produced by repeated immunizations with Human Haptoglobin protein.
Specificity
Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum. Analysis by SDS-PAGE was used to determine that this preparation is substantially free of aggregates and shows a banding pattern consistent with purified Rabbit IgG.
Applications
  • IHC
  • IHC - Paraffin (0.31 µg/ml)
  • Western blot (1:2000 - 1:10000)
  • ELISA (1:500000)

Usage
Immunohistochemistry: LS-B32 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B32 was determined to be 0.31 ug/ml.
Presentation
0.02 M Potassium Phosphate, pH 7.2, 0.15 M NaCl, 0.01% Sodium Azide
