GLUP / PACRG Antibody - Anti-PACRG Antibody - Western Blot. Western blot of Affinity Purified anti-PACRG antibody shows detection of a band ~40 kD corresponding to human PACRG (arrowhead lane 1). Specific reactivity with this band is blocked when the antibody is pre-incubated with the immunizing peptide (lane 2). Approximately 35 ug of a mouse embryonic fibroblast (MEF) whole cell lysate was separated by 4-20% SDS-PAGE and transferred onto nitrocellulose. After blocking the membrane was probed with the primary antibody diluted to 1:1500 for 2h at room temperature followed by washes and reaction with a 1:10000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX ( for 45 min at room temperature. IRDye800 fluorescence image was captured using the Odyssey Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.

Description
GLUP (PACRG) is a protein that is conserved across metazoans. In vertebrates, this gene is linked in a head-to-head arrangement with the adjacent parkin gene, which is associated with autosomal recessive juvenile Parkinson's disease. These genes are co-regulated in various tissues and they share a bi-directional promoter. Both genes are associated with susceptibility to leprosy. The parkin co-regulated gene protein forms a large molecular complex with chaperones, including heat shock proteins 70 and 90, and chaperonin components. This protein is also a component of Lewy bodies in Parkinson's disease patients, and it suppresses unfolded Pael receptor-induced neuronal cell death. Multiple transcript variants encoding different isoforms have been found for this gene. References: The UniProt Consortium. This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to an internal region of Human PACRG protein.
Specificity
A BLAST analysis was used to suggest reactivity with this protein from human, mouse and chicken based on 100% homology for the immunogen sequence. Cross reactivity with PACRG protein from zebrafish and rat may occur as this sequence only varies by one amino acid residue (94% homology). Expect reactivity with both splice variants of this protein. Cross reactivity with PACRG homologues from other sources has not been determined.

Usage
Immunohistochemistry: LS-B127 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B127 was determined to be 2.5 ug/ml.
