Description
CD163 (also called scavenger receptor cysteine-rich type 1 protein M130) is an acute-phase–regulated receptor that is involved in the clearance and endocytosis of hemoglobin/haptoglobin complexes by macrophages. It has a plasma membrane-bound form and a soluble form. CD163 is expressed on monocytes and mature resident macrophages such as red pulp macrophages in the spleen, Kupffer cells in the liver, alveolar and interstitial macrophages in the lung, medullary and perifollicular macrophages in lymph nodes, and perivascular and meningeal macrophages in the brain. The soluble form may be a biomarker for inflammatory diseases. Antibodies to CD163 can be used to identify monocytes and macrophages. It is positive in histiocytic sarcomas. Usage
Immunohistochemistry: LS-B3781 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B3781 was determined to be 20 ug/ml. Tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at less than or equal to 0.5 ug per test. A test is defined as the amount (ug) of antibody that will stain a cell sample in a final volume of 100 ul. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
