Description
Annexin A1 (ANXA1, lipocortin 1) is a calcium-dependent, phospholipid-binding protein that inhibits phospholipase A2 and exerts anti-inflammatory activity. It is expressed in tissues such as the bone marrow and the immune system, lung, and the gastrointestinal tract and is localized to the cytoplasm, plasma membrane, and nucleus. Dysregulation of ANXA1 expression and changes in subcellular localization are associated with many cancers, some with increased expression and some with decreased expression depending on the tissue and state of disease progression. Staining is used to distinguish hairy cell leukemia from other B-cell malignancies. In normal tissues, ANXA1 stains positive in squamous epithelium of the esophagus, tonsil, placental trophoblasts, and infiltrating leukocytes across a variety of tissues. Usage
Dilute antibody to its recommended working dilution using the same buffer defined in the Presentation. Manual IHC Protocol: 1) Perform Heat-Induced Epitope Retrieval at pH 9 for 10 to 30 minutes. 2) Block in peroxidase blocking solution for 5 minutes at room temperature. (Not required if using Alkaline Phosphatase System) 3) Dilute antibody 1:100-1:200 before applying. Incubate antibody for 10 to 30 minutes at room temperature. 4) Add secondary antibody and incubate for 20 to 30 minutes at room temperature. 5) Add DAB or Fast Red and incubate for 5 to 10 minutes at room temperature. 6) Counterstain with hematoxylin for 0.5 to 5 minutes, depending on the hematoxylin used. Rinse with distilled water and blueing solution for 30 seconds. Dehydrate and apply coverslip.
