Immunohistochemistry Services

Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

TCR Screening Services

Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Specifications

Description
DPP4 is a cell surface glycoprotein receptor that functions in immune regulation, apoptosis, signal transduction, glucose metabolism and regulation of insulin secretion. It can inactivate GLP-1, PACAP and GIP, which each stimulate insulin secretion in the pancreas and reduce glucose levels. It acts as a positive regulator of T-cell coactivation and induces T-cell proliferation and NF-kappa-B activation, and it is involved in lymphocyte-epithelial cell adhesion. In disease, DPP4 contributes to fibrosis in the liver, kidney and other organs, and it is also deregulated in a number of cancers. Furthermore, it is a target of therapies in diabetes. In immunohistochemistry, DPP4 has luminal membranous positivity in the parathyroid, small intestine, kidney, endometrium, placenta and prostate. Immunogen
Synthetic 15 amino acid peptide from N-Terminus of human CD26. Percent identity with other species by BLAST analysis: Human, Orangutan, Gibbon, Monkey (100%); Gorilla, Marmoset, Bovine (93%); Cat, Rabbit (87%); Dog, Bat, Elephant, Panda, Horse (80%).
Blocking Peptide
LS-E29323 - Liquid - 50 µg - $145.00
Immunizing peptide used to generate LS-A8059. Useful for pre-absorption and neutralization of the antibody's antigen binding site.
Epitope
N-Terminus
Specificity
Human CD26. BLAST analysis of the peptide immunogen showed no homology with other human proteins.
Usage
Immunohistochemistry: The unconjugated form was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for the unconjugated form was determined to be 2.5 ug/ml.
