This ELISA kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with Pros1. During the reaction, Pros1 in the sample or standard competes with a fixed amount of Pros1 on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to Pros1. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin(SABC) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of Pros1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

PROS1 / Protein S is a vitamin K-dependent plasma protein that functions as a cofactor for the anticoagulant protease, activated protein C (APC) to inhibit blood coagulation. It is found in plasma in both a free, functionally active form and also in an inactive form complexed with C4b-binding protein. Mutations in this gene result in autosomal dominant hereditary thrombophilia. An inactive pseudogene of this locus is located at an adjacent region on chromosome 3.
