LS-F36732 is a 96-well enzyme-linked immunosorbent assay (ELISA) for the Quantitative detection of Mouse F9 / Factor IX in samples of Plasma, Serum and Tissue Homogenates. It is based upon a Sandwich assay principle and can be used to detect levels of F9 / Factor IX as low as 0.094 nanograms per millilter.

This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- FIX antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-FIX antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the FIX amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration ofFIX can be calculated.
F9 / Factor IX is a vitamin K-dependent coagulation factor IX that circulates in the blood as an inactive zymogen. This factor is converted to an active form by factor XIa, which excises the activation peptide and thus generates a heavy chain and a light chain held together by one or more disulfide bonds. The role of this activated factor IX in the blood coagulation cascade is to activate factor X to its active form through interactions with Ca+2 ions, membrane phospholipids, and factor VIII.
