LS-F39414 is a 96-well enzyme-linked immunosorbent assay (ELISA) for the Quantitative detection of Human GHRL / Ghrelin Preproprotein in samples of Plasma, Serum and Tissue Homogenates. It is based upon a Competitive EIA assay principle and can be used to detect levels of GHRL / Ghrelin Preproprotein as low as 28.125 picograms per milliliter.
About GHRL / Ghrelin Preproprotein
GHRL / Ghrelin Preproprotein is the ghrelin-obestatin preproprotein that is cleaved to yield two peptides, ghrelin and obestatin. Ghrelin is a powerful appetite stimulant and plays an important role in energy homeostasis. Its secretion is initiated when the stomach is empty, whereupon it binds to the growth hormone secretagogue receptor in the hypothalamus which results in the secretion of growth hormone (somatotropin). Upon receipt of an order each kit is assembled and tested to ensure that it meets specifications before shipping. Minor changes may occur to the Range, Sensitivity, and Precision. In the event of a significant change the order would be confirmed with the customer before shipping ELISA kit lot numbers reflect the date of final assembly and testing for each specific kit rather than a bulk manufactured lot. All kits are tested to confirm that they fall within their defined Inter- and Intra- assay coefficient of variation.

Background
This ELISA kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with GHRL. During the reaction, GHRL in the sample or standard competes with a fixed amount of GHRL on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to GHRL. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin(SABC) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of GHRL in the samples is then determined by comparing the O.D. of the samples to the standard curve.
