Description: The transduction of neurobehavioral effects by serotonin (5-hydroxy tryptamine; 5-HT) is mediated via at least six major 5-HT receptor subtypes, including 5-HT2 AL 34662 is a potent 5-HT2 receptor agonist with ocular hypotensive activity. It binds to the human and rat 5-HT2 receptors in cerebral cortex homogenates with IC50 values of 1.5 and 0.77 nM, respectively. AL 34662 binds to the recombinant human 5-HT2A, 5-HT2B, and 5-HT2C receptors with IC50 values of 14.5, 8.1, and 3 nM, respectively. In ocular hypertensive cynomolgus monkey eyes, AL 34662 lowered intraocular pressure (IOP) 25% at a dose of 100 ug and 33% at 300 ug (six hours post dose) with minimal side effects.
AL 34662 is supplied as a crystalline solid. A stock solution may be made by dissolving the AL 34662 in an organic solvent purged with an inert gas. AL 34662 is soluble in organic solvents such as ethanol, DMSO, and dimethyl formamide (DMF). The solubility of AL 34662 in ethanol is approximately 1 mg/ml and approximately 10 mg/ml in DMSO and DMF. AL 34662 is sparingly soluble in aqueous buffers. For maximum solubility in aqueous buffers, AL 34662 should first be dissolved in DMSO and then diluted with the aqueous buffer of choice. AL 34662 has a solubility of approximately 0.2 mg/ml in a 1:4 solution of DMSO:PBS (pH 7.2) using this method. We do not recommend storing the aqueous solution for more than one day.
