Description: Angiotensin II is a hormone that plays an important role in regulating blood pressure. Elevated levels of angiotensin II are implicated in inducing and maintaining hypertension, and also in the development of atherosclerosis. Both of these effects are mediated by the angiotensin II type 1 (AT1) receptor. Losartan is an AT1 receptor antagonist with a Ki value of 5-20 nM. It has an attenuating effect on vein graft atherosclerosis in rabbits and effectively reduces arterial blood pressure in rats. In humans, losartan controls hypertension while protecting renal function.

Losartan (potassium salt) is supplied as a crystalline solid. A stock solution may be made by dissolving the losartan (potassium salt) in an organic solvent purged with an inert gas. Losartan (potassium salt) is soluble in organic solvents such as ethanol, DMSO, and dimethyl formamide. The solubility of losartan (potassium salt) in these solvents is at least 20 mg/ml. Further dilutions of the stock solution into aqueous buffers or isotonic saline should be made prior to performing biological experiments. Ensure that the residual amount of organic solvent is insignificant, since organic solvents may have physiological effects at low concentrations. Organic solvent-free aqueous solutions of losartan (potassium salt) can be prepared by directly dissolving the crystalline compound in aqueous buffers. The solubility of losartan (potassium salt) in PBS (pH 7.2) is at least 10 mg/ml. We do not recommend storing the aqueous solution for more than one day.
