MEKK2, a MAP3K type protein kinase, is a member of MAP kinase signaling pathways. The protein contains a C-terminal protein kinase domain and a non-catalytic N-terminal domain. MEKK2 has been shown to regulate both JNK and ERK cascades by phosphorylating and activating either MKK4/MAP2K4 or MEK1/MAP2K1, respectively. Additionally, MEKK2 has been shown to directly phosphorylate and activate Ikappa B kinases (IKKs) of the immune and inflammatory response NF-kappa B signaling pathway and to bind and activate protein kinase C-related kinase 2. MEKK2 (-/-) embryonic stem cell-derived mast cells demonstrate that this protein is required for cytokine production and receptor-mediated stimulation of JNK.
