The reactivity of the antiserum is restricted to the Fc portion of the IgD molecule, which represents strict isotype (class specificity. In immunoelectrophoresis and radial immunodiffusion, using various antiserum concentrations against mouse serum belonging to different allotypic groups, a single characteristic precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with the purified IgD from Balb/C and C57Bl. No precipitation reaction is obtained with purified IgG, including all subclasses, IgA, IgM, IgG/Fab fragments or other serum proteins. The antiserum also reacts with membranebound IgD in peripheral blood cells of different mouse strains as tested by immunofluorescence microscopy. In precipitating techniques as immunoelectrophoresis and radial immunodiffusion to identify the presence of IgD in mouse serum and other body fluids or to determine its concentration.
