growth hormone releasing hormone receptor

GHRHR, a Growth Hormone-Releasing Hormone Receptor, is involved in the proliferation and differentiation of somatotrophic pituitary cells and in disease. The GHRH receptor stimulates growth hormone (GH) secretion and synthesis in the pituitary. Mutations in this receptor cause either growth-hormone deficiency, resulting in dwarfism, or excessive production of the hormone, leading to gigantism. Four alternative splice variants have been identified that are tissue specific.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0) . Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4 °C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

GHRHR Antibody - Immunofluorescent analysis of GHRH Receptor staining in LOVO cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
