CDC27 shares strong similarity with Saccharomyces cerevisiae protein Cdc27, and the gene product of Schizosaccharomyces pombe nuc 2. This protein is a component of the anaphase-promoting complex (APC), which is composed of eight protein subunits and is highly conserved in eukaryotic cells. This complex catalyzes the formation of cyclin B-ubiquitin conjugate, which is responsible for the ubiquitin-mediated proteolysis of B-type cyclins. The protein encoded by this gene and three other members of the APC complex contain tetratricopeptide (TPR) repeats, which are important for protein-protein interactions. This protein was shown to interact with mitotic checkpoint proteins including Mad2, p55CDC and BUBR1, and it may thus be involved in controlling the timing of mitosis. Alternative splicing of this gene results in multiple transcript variants. Related pseudogenes have been identified on chromosomes 2, 22 and Y.

![CDC27 Antibody - Anti-cdc27 Antibody - Western Blot. Western blot of Affinity Purified anti-cdc27 antibody shows detection of a band ~90 kD corresponding to human cdc27 (arrowhead). Approximately 35 ug of HeLa whole cell lysate was separated by SDS-PAGE and transferred onto nitrocellulose. After blocking the membrane was probed with the primary antibody diluted to 1.0 ug/ml for 2 h at room temperature followed by washes and reaction with a 1:10000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX ( for 45 min at room temperature. IRDye800 fluorescence image was captured using the Odyssey Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Lane 2 shows lysate from cells treated with nocodazole. Phosphorylated CDC27 is mostly present only in cell preparations arrested in mitosis. Each lane contains approximately 30 ug of HeLa whole cell lysates separated by 12.5% SDS-PAGE followed by transfer to nitrocellulose. After blocking with 5% non-fat dry milk in TTBS, the membrane was probed with the primary antibody diluted to 1:500 for 1 h at room temperature followed by washes and reaction with a 1:5000 dilution of HRP Gt-a-Rabbit IgG [H&L] MX (LS-C60865) for 45 min at room temperature. ECL reagent was used for detection. Other detection systems will yield similar results.
