Specificity and Use

WASL / N-WASP antibody was raised against e. coli-derived rhN-WASP (aa21-270).
Selected for its ability to recognize human N-WASP in the applications listed.
Western Blot: This antibody can be used at 1.0 ug/ml with the appropriate secondary reagents to detect N-WASP. Immunocytochemistry: This antibody has been used at a concentration of 10 ug/ml to detect human N-WASP on A431 human epidermoid carcinoma cells stimulated with EGF. Cells were fixed with PBS containing 4% paraformaldehyde and blocked with PBS containing 10% normal donkey serum, 0.1% Triton X-100, and 1% BSA. After blocking, cells were incubated with diluted primary antibody followed by Rhodamine RedTM-coupled anti-goat IgG at room temperature in the dark. Between each step, cells were washed with PBS containing BSA. Direct ELISA: This antibody can be used at 0.5-1.0 ug/ml with the appropriate secondary reagents to detect human N-WASP. The detection limit for rhN-WASP is approximately 1.0 ng/well.

WASL / N-WASP is a member of the Wiskott-Aldrich syndrome (WAS) protein family. Wiskott-Aldrich syndrome proteins share similar domain structure, and associate with a variety of signaling molecules to alter the actin cytoskeleton. The encoded protein is highly expressed in neural tissues, and interacts with several proteins involved in cytoskeletal organization, including cell division control protein 42 (CDC42) and the actin-related protein-2/3 (ARP2/3) complex.
