Specificity and Use

VEGFA / VEGF antibody was raised against this purified antibody was prepared from whole rabbit serum produced by repeated immunizations with full length recombinant human VEGF-165 protein.
This purified antibody has been heated to 56°C for 30 minutes. In ELISA and other immunoreactive assays, this antibody will recognize both native and recombinant human VEGF-165 in cell supernatants and certain body fluids. A control of similarly diluted normal rabbit IgG is recommended.
This purified antibody has been tested for use in ELISA and western blotting. Reactivity is also expected in neutralizations, radioimmunoassay and immunohistochemistry. The endotoxin content is estimated to be <10 pg/ul by the LAL method. By western blot a band approximately 22 kD in size corresponding to native monomeric human VEGF-165 protein is expected in the appropriate cell lysate or extract. Specific conditions for reactivity should be optimized by the end user.

VEGFA / VEGF is a member of the PDGF/VEGF growth factor family and encodes a protein that is often found as a disulfide linked homodimer. This protein is a glycosylated mitogen that specifically acts on endothelial cells and has various effects, including mediating increased vascular permeability, inducing angiogenesis, vasculogenesis and endothelial cell growth, promoting cell migration, and inhibiting apoptosis.

Anti-VEGF-165 Antibody - Western Blot. anti-Human VEGF-165 antibody shows detection of a band ~22 kD in size corresponding to recombinant human VEGF-165. This isoform of VEGF tends to form a disulfide linked homodimer. Molecular weight markers are also shown (left). After transfer, the membrane was blocked overnight with 3% BSA in TBS followed by reaction with primary antibody at a 1:1000 dilution. Detection occurred using peroxidase conjugated anti-Rabbit IgG (LS-C60865) secondary antibody diluted 1:40000 in blocking buffer (p/n MB-070) for 30 min at RT followed by reaction with FemtoMax chemiluminescent substrate. Image was captured using VersaDoc MP 4000 imaging system (Bio-Rad).
