  • Immunohistochemistry Services

  • Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

  • TCR Screening Services

  • Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Specificity and Use

VCAN / Versican antibody was raised against recombinant protein corresponding to aa1344-1554 from human Versican isoform V0, expressed in E. coli (P13611).
Recognizes human Versican. Species sequence Homology: For immunizing sequence, aa1344-1554: canine (80%), mouse (60%).
Suitable for use in ELISA, Western Blot, Immunocytochemistry and Immunohistochemistry. Western Blot: 0.1-0.2 ug/ml. Detection limit for rhVersican is ~2 ng/lane under non-reducing and reducing conditions. Immunocytochemistry: 10 ug/ml detects Versican in Hela cells. Cells were fixed with PBS containing 4% paraformaldehyde for 20 minutes at RT and blocked with PBS containing 10% normal donkey serum, 0.1% Triton X-100, and 1% BSA for 45 minutes at RT. After blocking, cells were incubated with diluted primary antibody overnight at 4? C followed by fluorochrome-coupled anti-goat IgG at RT in the dark for one hour. Between each step, cells were washed with PBS containing 0.1% BSA. Immunohistochemistry: 10 ug/ml detects Versican in cryosections of E13.5 mouse embryos. Tissue was fixed with PBS containing 4%PFA for 20 min. at RT and blocked with PBS containing 10% normal donkey serum, 0.1% Triton and 1% BSA for 45 minutes at RT. After blocking, sections were incubated with diluted primary antibody for 3 hours at RT followed by fluorochrome-coupled anti-goat lgG at RT for 1 hour. Between each step, sections were washed with 0.1% BSA in PBS. Direct ELISA: 0.5-1.0 ug/ml. Detection limit for rhVersican is ~0.1 ng/well. Optimal dilutions should be determined by each laboratory.

Lyophilized from PBS, 5% trehalose.
500 µl Sterile PBS

VCAN / Versican is a member of the aggrecan/versican proteoglycan family. The protein encoded is a large chondroitin sulfate proteoglycan and is a major component of the extracellular matrix. This protein is involved in cell adhesion, proliferation, proliferation, migration and angiogenesis and plays a central role in tissue morphogenesis and maintenance. Mutations in this gene are the cause of Wagner syndrome type 1.
