Specificity and Use

TPSAB1 / Mast Cell Tryptase antibody was raised against nS0-derived, recombinant murine Tryptase beta1 (aa29-273).

Recognizes murine Tryptase, beta1 in direct ELISAs and Western Blots. Shows ~10% cross-reactivity with recombinant human TPSB2, 5% cross-reactivity with recombinant human TPS1 and less than 1% cross-reactivity with recombinant human TPSG1 and recombinant murine MCPT1.
Suitable for use in ELISA, Western Blot and Immunoprecipitation. Direct ELISA: 0.5-1 ug/ml. The detection limit for recombinant murine Tryptase beta1 is ~0.5 ng/well. Western Blot: 0.1-0.2 ug/ml. The detection limit for recombinant murine Tryptase beta1 is ~1 ng/lane under non-reducing and reducing conditions. Immunoprecipitation: Immunoprecipitates recombinant, murine Tryptase beta1 from conditioned media of transfected NS0 cells. The recovered recombinant murine Tryptase beta1 can be detected by Western Blot analysis using a monoclonal antibody.

Tryptases comprise a family of trypsin-like serine proteases, the peptidase family S1. Tryptases are enzymatically active only as heparin-stabilized tetramers, and they are resistant to all known endogenous proteinase inhibitors. Several tryptase genes are clustered on chromosome 16p13.3. These genes are characterized by several distinct features.
