TNF Alpha is a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. This cytokine is mainly secreted by macrophages. It can bind to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. This cytokine is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
TNF alpha in Human PBMCs. TNF alpha was detected in immersion fixed human peripheral blood mononuclear cell s (PBMCs) stimulated with LPS and monensin using Mouse Anti-Human TNF alpha Monoclonal Antibody at 10 ug/ml for 3 hours at room temperature. Cells were stained using NorthernLights 557-conjugated Anti-Mouse IgG Secondary Antibody (yellow) and counterstained with DAPI (blue).
Cytotoxicity Induced by TNF- alpha and Neutralization by Human TNF- alpha Antibody. Recombinant Human TNF- alpha induces cytotoxicity in the the L-929 mouse fibroblast cell line in a dose-dependent manner (orange line). Cytotoxicity elicited by Recombinant Human TNF- alpha (0.25 ng/ml) is neutralized (green line) by increasing concentrations of Mouse Anti-Human TNF- alpha Monoclonal Antibody. The ND50 is typically 0.04-0.08 ug/ml in the presence of the metabolic inhibitor actinomycin D (1 ug/ml).
