Specificity and Use

TLR4 antibody was raised against human TLR4
This HTA125 antibody has been pre-titrated and tested by flow cytometric analysis of human peripheral blood leukocytes. This can be used at 5 ul (1 ug)/per test. A test is defined as the amount (ug)/test of antibody that will stain a cell sample in a final volume of 100 ul. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is highly recommended that for optimal staining of TLR4, whole blood be stained using the LWB protocol rather than Ficoll-gradient prepared PBMCs. The use of a density gradient appears to reduce the staining intensity significantly. Important Note: Human TLR4 is expressed very dimly on the surface of peripheral blood monocytes from normal donors. For best detection of TLR4 on these cells, LS-C105972, PE-conjugated HTA125, is recommended. APC and FITC conjugated HTA125 are useful for detection of surface human TLR4 on cell lines/transfected cells that have high expression levels. The applications listed have been tested for the unconjugated form of this product. Other forms have not been tested.

Cooperates with LY96 and CD14 to mediate the innate immune response to bacterial lipopolysaccharide (LPS). Acts via MYD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Also involved in LPS-independent inflammatory responses triggered by free fatty acids, such as palmitate, and Ni2+. Responses triggered by Ni2+ require non-conserved histidines and are, therefore, species-specific.
