Specificity and Use

TLR4 antibody was raised against synthetic peptide from human TLR4.
aa100-200
The amino acid sequence used has 100% identity in human, chimp, baboon, Chinese hamster, and rat; 93% identity in bovine and pig; 85% identity in sheep and 78% identity in cat, horse and mouse.
Immunohistochemistry: LS-B2070 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2070 was determined to be 5 ug/ml.

Cooperates with LY96 and CD14 to mediate the innate immune response to bacterial lipopolysaccharide (LPS). Acts via MYD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Also involved in LPS-independent inflammatory responses triggered by free fatty acids, such as palmitate, and Ni2+. Responses triggered by Ni2+ require non-conserved histidines and are, therefore, species-specific.
