Specificity and Use

Purified canine TSH.
Polyclonal antibody was raised against highly purified intact canine thyroid stimulating hormone (TSH) and is specific for canine TSH. Exhibits less than 0.1% cross reactivity with the other major canine glycoprotein hormones; growth hormone (GH), luteinizing hormone (LH) and follicle stimulating hormone (FSH) or prolactin using an iodinated hormone tracer assay. This strongly indicates thatantibody is primarily reactive with the TSHbeta chain although it may also have reactivity with beta chain conformational epitopes dependent on the presence of the common alpha chain shared with the other glycoprotein hormones. TSH, also known as thyrotropin is synthesized in, and secreted from within the anterior pituitary gland, under the influence of secreted thyrotropin releasing hormone from the hypothalamus. TSH is composed of, as are the other major glycoprotein hormones, a common alpha chain non-covalently linked to a unique beta chain responsible for conferring the structural and biological identity of the individual hormone. TSH binds to the TSH receptor (TSHR) in the thyroid gland. TSHR has also bee detected in other cell types including adipocytes, neuronal cells and gastric mucosa. Defects in the TSHbeta subunit can lead to hypothyroidism, either through the lack of secreted hormone or secretion of TSH lacking full biological acticvity.
