Specificity and Use

TH / Tyrosine Hydroxylase antibody was raised against denatured tyrosine hydroxylase from rat pheochromocytoma (denatured by sodium dodecyl sulfate).
Recognizes tyrosine hydroxylase. Species cross-reactivity: Mouse, human, ferret, feline and Aplysia (marine mollusc). Because Tyrosine Hydroxylase is highly conserved, this antibody is expected to cross-react with most mammalian species and many non-mammalian species.
Immunohistochemistry: LS-B3443 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. Western Blot: 1:1000 on 10 ug of PC12 lysates. Immunofluorescence: 1:100-1:1000. Fixed with 4% PFA overnight at 4¦C.

Tyrosine hydroxylase in involved in the conversion of phenylalanine to dopamine. It is the rate-limiting enzyme in the synthesis of catecholamines and as such has a key role in the physiology of adrenergic neurons. TH is encoded by four distinct mRNAs produced by alternative splicing of a single primary transcript. Expression of the mRNAs varies in different parts of the nervous system. Defects in TH are the cause of autosomal recessive Segawa syndrome.
