Specificity and Use

TCF7 antibody was raised against synthetic peptide (KLH-coupled) corresponding to a region surrounding Pro95 of human TCF1 protein.
Detects endogenous levels of total TCF1 protein. Does not recognize the dominant negative isoforms of TCF1 lacking the amino-terminal beta-catenin binding domain and does not cross-react with LEF1.
Suitable for use in Immunofluorescence, Flow Cytometry, Western Blot, Immunoprecipitation, Immunohistochemistry and Immunocytochemistry. Flow Cytometry: 1:200. Immunocytochemistry/Immunofluorescence: 1:25. Immunohistochemistry (paraffin): 1:100. Immunoprecipitation: 1:50. Western Blot: 1:1000.


10 mM sodium HEPES, pH 7.5, 150 mM sodium chloride, 100 ug/ml BSA, 50% glycerol, less than 0.02% sodium azide.

Transcriptional activator involved in T-cell lymphocyte differentiation. Necessary for the survival of CD4+ CD8+ immature thymocytes. Isoforms lacking the N-terminal CTNNB1 binding domain cannot fulfill this role. Binds to the T-lymphocyte-specific enhancer element (5'-WWCAAAG-3') found in the promoter of the CD3E gene. May also act as feedback transcriptional repressor of CTNNB1 and TCF7L2 target genes. TLE1, TLE2, TLE3 and TLE4 repress transactivation mediated by TCF7 and CTNNB1.
