Specificity
The antibody is specific for Synaptophysin. The specificity was ascertained by immunoblotting and immunohistochemistry. The antibody predominantly reacts with a 38 kD transmembrane glycoprotein from synaptic vesicles. Antigen was isolated from presynaptic vesicles. Antigen was isolated from presynaptic vesicles. Usage
Staining of normal neuroendocrine and neoplastic neuroendocrine cells in tissue sections. The antibody recognizes presynaptic vesicles of neurons of brain, spinal cord, retina neuromuscular junctions, small vesicles of adrenal medulla and pancreatic islets. The antibody can be used for the characterization of neuroendocrine tumors. The antibody can be used on frozen sections and paraffin embedded tissue, proteolytic or T. U. F. pretreatment is not necessary but may, depending on the length of the fixation period, enhance the staining. As positive control adrenal pheochromocytoma and pancreatic islet cell neoplasm are recommended. A dilution of 1:10 is recommended, based on the Indirect method, with a normal ABC method the dilutions can be 5-10x higher, but the optimal dilution should be tested by serial dilution.
