Specificity and Use

SPTAN1 / Alpha Fodrin antibody was raised against recombinant construct containing the 13th, 14th and 15th spectrin repeats of hiuman α-II spectrin (amino acids 1398-1685). This construct was expressed in and purified from E. coli.
Human SPTAN1 / Alpha Fodrin
The antibody preparation can be diluted to 1:500-1:1000 for immunofluorescence staining and 1:5000-1:10000 for western blotting. On western blots look for a major band at 240 kDa, depending on the species.

Spectrins are a family of filamentous cytoskeletal proteins that function as essential scaffold proteins that stabilize the plasma membrane and organize intracellular organelles. Spectrins are composed of alpha and beta dimers that associate to form tetramers linked in a head-to-head arrangement. This gene encodes an alpha spectrin that is specifically expressed in nonerythrocytic cells.

The alpha-II spectrin antibody stains numerous axonal and dendritic profiles in these cultures, and this image shows an optical section through a group of neuronal cell bodies. SPTAN1 / Alpha Fodrin antibody clearly reveals the submembranous cytoskeleton. Since alpha-II spectrin is specific for neurons in the CNS, the glial cells in this culture are not recognized by this antibody. This antibody also reveals the submembranous cytoskeleton of the axon.

Western blots of extract of bovine spinal cord probed with SPTAN1 / Alpha Fodrin antibody. A prominent band running at 240 kDa represents the intact alpha-II spectrin heavy chain. More minor bands below this likely represent in vivo proteolytic fragments of alpha-II spectrin.
