Specificity and Use

SPP1 / Osteopontin antibody was raised against synthetic peptide from human SPP1 / Osteopontin.
Synthetic peptide C-K-S-K-K-F-R-R-P-D-I-Q-Y-P-D corresponding to human osteopontin proteins conjugated to KLH using maleimide. A residue of cysteine was added to the amino terminal end to facilitate coupling.
Immunohistochemistry: LS-B425 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B425 was determined to be 1:500.

Binds tightly to hydroxyapatite. Appears to form an integral part of the mineralized matrix. Probably important to cell-matrix interaction.Acts as a cytokine involved in enhancing production of interferon-gamma and interleukin-12 and reducing production of interleukin-10 and is essential in the pathway that leads to type I immunity.


Rabbit anti-Osteopontin was used at a 1:100-1:300 dilution to detect osteopontin by immunohistochemistry. Osteopontin is a normal component of elastic fibers in the breast (shown heavily stained in this section of human breast tumor cells). There is also weak staining of the extracellular matrix. Osteopontin is not expressed in breast tumor cells, and there is no staining of the breast cells in this section. No antigen retrieval is required.
